Chinical Studies on Immunological Pregnancy Test by Slide Method


Abstract
From clinical studies on immunological diagnostic pregnanoy test with slide method for diagnostic and follow up evaluations in norma1 pregnancy, hydatidiformmole, choriocarcinoma and others, the following results were obtained.
1. The positive HCG test was able to read within 2 minutes using 1 drop (0.07ml) of the urine to he tested and all urines from normal nonpregnant women, normal healthy men and saline revealed negative results.
2. The rate of positivity was 96.6% in 89 cases of normal pregnancy and 3 cases of negativity were noted among first and second month of the pregnancy.
3. The highest quantity of HCG was estimated at the third month of the pregnancy. i.e 16 fold dilution (91,424 I.U.) of urine exhibited positive result.
4. The positivity of HCG test was increased by Zondek¢¥s alcoholic concentration method with pregnant urine. Two of them were resulted in positive conversion in 2 fold dilution by this method from negative HCG test in undiluted urine and another one was increased the positivity to 6 fold dilution from 4 fold, and two were 25 fold dilution from 16 fold.
5. The negative conversion of HCG test was initiated approximately from the fifth day of postpartum.
6. The minimally positive limit of the reaction on this slide test was 0.4 I.U. per drop of the urine to be tested by using standard HCG preparation (500 I.U.).
7. No specific influence to the storage of the urine to be tested in the refrigerator (4¡É) was observed during 3weeks.
8. On the relations between temperature and HCG slide test. the reaction time was shortened as the temperature became high and as the temperature became low, the reaction was delayed. But it was most reliable to read the reaction at the room temperature.
9. No influence to the pH of urine(5.5¡­7.0) on HCG test was boserved.
10. In the influence of the protein in the urine from non pregnant which was negative HCG test, the protein (0.2% or more) induced false positive results.
11. The rate of positivity of choriocarcinorma was 100% and was 98.2% in hydatidiform mole.
12. There was remarkable change in progress of choriocarcinoma with methotrexate therapy and in most of hydatidiform mole the negative conversion of HCG test was noted after evacuation.
